Much as a troublehooting guide is useful I do wonder whether or not this is appropriate for an encyclopedia, perhaps it could be labelled as common problems / solutions?

Suggested addition: bead beating under the lysis step

Disclosure: I am associated with LabTIE International, a manufacturer of laboratory bead dispensing equipment (see my user page), so I am proposing this change here rather than editing the article directly. The proposed text cites only independent literature and does not mention or link my company.

The article currently notes that "mechanical or enzymatic methods" are often used for lysis, but gives no detail on mechanical lysis. I suggest adding something like the following to the lysis step:

"Mechanical lysis is commonly performed by bead beating, in which samples are shaken at high speed together with small abrasive beads (typically glass, ceramic, zirconium or steel), releasing DNA from cells that resist chemical or enzymatic lysis, such as mycobacteria, spores and plant tissue.[1] In large-scale studies, extraction is commonly performed in 96-well plate format so that many samples can be processed in parallel, although this format requires precautions against well-to-well contamination.[2]"
