DNA-guided CRISPR

Modified versions of Class II systems (specifically Cas12a) function with an artificial single-stranded DNA guide in place of natural RNA.^[1] Within this new paradigm, the system's endonuclease activity now is redirected towards cleaving ssRNA instead of dsDNA.^[2]^[3] With DNA as the guide, the need for intricate RNA preparation and preservation is no longer required, allowing Class II effectors more versatility in programmable RNA targeting and translational biotechnology.^[2]^[4] EliStewart43 (talk) 03:09, 31 May 2026 (UTC)Reply
