For all the poor/meaningless nomenclature, Fas is yet the name of the receptor. The gene that encodes the receptor is TNFRSF6, and thus fas is the processed TNFRSF6 gene product. The literature still pretty much to the terminology Fas and Fas Ligand (FasL).

So what is missing in the article.

1. FADD has a death domain, which allows it to interact with other proteins that have death domains like TRADD, and the death domains of TNF-Recpetor like proteins with appropriate death domains. Examples are Fas, TRADD, etc.

2. FADD has a death effector domain (DED) which allows it to interact with other proteins with death effector domains. Examples, Caspase-8, Caspase-10.


So what is needed is a far less complicated graphic illustrating the -DD-DED- structure of FADD.

I should also add the figure legend does not show TNF R1 activation pathway, that is TNFRSF1A. Confusing, I know butthere are 30 or so different TNFRSF and the one that is displayed is the FasL/Fas apoptosis pathway. PB666 yap 02:37, 30 April 2011 (UTC)Reply
