Notes on the structure of Adamax
[edit]There seems to be some confusion around the structure of Adamax. It is described as being an analogue of Semax but with the N- and C- terminal modifications found in Peptide 021, i.e. where semax is MEHFPGP, adamax is Ac-MEHFPGP-AG-NH2.
The structure of semax is well established;
while the structure of Peptide 021 has also been published;
and by applying these modifications to semax we can derive the structure of Adamax;
However, many commercial suppliers of what is claimed to be "Adamax" seem not to have read the papers describing Peptide 021. The structures depicted by peptide manufacturers as Adamax fall into two groups. First is Ac-MEHFPGP-AG where "AG" has its conventional meaning of alanine-glycine, even though the websites selling this compound always describe it as having an adamantyl group, yet no adamantyl group is present in this structure;
while the second structure is closer as it does validly represent Ac-MEHFPGP-AG-NH2, and contains an adamantyl group, however in this case "AG" has been interpreted to mean the conventional meaning of adamantyl glycine, which is to say an amino acid like alanine but with the CH3 replaced by adamantyl, rather than the unnatural adamantane containing amino acid found in Peptide 021;
Since most vendors of peptides to end users do not supply structure images at all, there is no way of knowing which of these three options is being supplied as "Adamax", but fortunately the three possible options all have different molecular weights and so can be readily distinguished by GC-MS. However, since all bulk peptide suppliers as of 2025 appear to list either structures 2 or 3, it can be concluded that almost none of what is currently sold as "Adamax" is the same compound sold by the original manufacturer (who has since gone out of business).
